摘  要:目的利用红色荧光蛋白(RFP)基因建立原位骨肉瘤肺转移裸鼠模型,为体内监测骨肉瘤的生长及抗肿瘤药物的药效评价建立一种肿瘤动物模型。方法以包含逆转录病毒pLNCX-DsRed-2的PT67包装细胞转染人骨肉瘤细胞系143B,在裸鼠胫骨近端原位接种使其成瘤,活体荧光成像系统观察骨肉瘤的生长及肺转移情况。结果获得稳定表达红色荧光蛋白的人骨肉瘤细胞株,原位接种的肿瘤发生率达100%,肺转移率为80%。通过活体荧光成像系统于移植1周观察到肿瘤形成,移植4周肿瘤呈梭形膨胀,移植6周肿瘤呈分叶状球形,发光强度亦逐渐增加。结论通过建立原位骨肉瘤肺转移裸鼠模型可以观察原位骨肉瘤的不同生长过程以及肺转移情况,是研究骨肉瘤生长及肺转移相关因素的一种重要工具。Objective To establish an orthotopic osteosarcoma and pulmonary metastasis nude mice model expressing red fluorescent protein( RFP), and could be used to monitor the growth of osteosarcoma and evaluate the efficacy of antitumor drug. Methods Human osteosarcoma cells of 143B were transfected with incasing cells PT67 contained pLNCX-DsRed-2, RFP-tagged osteosarcoma cells were injected into the proximal tibia of athymic mice, the growth and pulmonary metastasis of osteosarcoma were observed and an- alyzed with whole body optical imaging system. Results The stable RFP tagged 143B cells were established and the cell line was used to duplicate the implanted tumor model, the incidence of orthotopic tumor was 100%, and 80% of them metastasized to lungs. The tumor was forming after implanting for 1 week, growing with a spindle shape after implanting for 4 weeks and with spherical shape for 6 weeks, luminous intensity enhancing gradually, could be observed by whole body optical imaging system. Conclusion The clinically relevant model of human osteosarcoma provides varying degrees of tumor growth at the primary site and lung metastatic potential.
