摘 要:目的探讨环氧化酶-2(COX-2)、骨桥蛋白(OPN)在人脑膜瘤中的表达及临床意义。方法选择脑膜瘤患者40例作为观察组,10例健康体检者作为对照组。采用ELISA法测定COX-2和OPN的浓度;采用免疫组化技术检测术后40例脑膜瘤组织及10例正常脑膜组织,COX-2、OPN及基质金属蛋白酶-9(MMP-9)的表达。结果侵袭组中COX-2、OPN的血清含量高于非侵袭组,差异有统计学意义(P〈0.01);同样COX-2、OPN及MMP-9蛋白在侵袭组脑膜瘤中的表达均高于非侵袭组(P〈0.01)。在侵袭性脑膜瘤中COX-2、OPN和MMP-9两两比较,其表达均具有显著相关性。结论COX-2、OPN与脑膜瘤的侵袭性密切相关,一定程度上可作为评价脑膜瘤侵袭性及预后的辅助指标,通过术前血清学检测、术后免疫组化检查可判断脑膜瘤的潜在侵袭性,指导临床治疗方案的制定,提高疗效水平。Objective To investigate the expression of cyclooxygenase-2 (COX-2) and osteopontin( OPN ) in meningioma and the clinical significance. Methods Forty cases of meningioma were choosed as observation group, and 10 cases of health people were choosed as control group. The serum concentrations of COX-2 and OPN were detected by enzyme linked immunosorbent assay. The expression levels of OPN, COX-2 and MMP-9 were detected by immunohistochemical S-P method in 40 cases of meningiomas and 10 cases normal brain tissue. Results The serum concertrations of OPN and COX-2 in invasive meningioma were higher than those in non-invasive ( P 〈 0. 01 ). The expression levels of COX- 2, OPN and MMP-9 in invasive meningioma was higher than that in non-invasive meningioma (P 〈 0. 01 ). There were significant relationps between COX-2, OPN and MMP-9 in invasive meningioma. Conclusions COX-2, OPN are closely correlated with meningioma invasiveness, indicating that COX- 2, OPN can evaluate invasiveness and prognostic of meningioma. Through detecting of COX-2, OPN in the samples of meningioma, we can judge the potential invasiveness of meningioma, and institute a right treatment plan to improve the curative effect of tumor patients.
