大孔吸附树脂分离纯化芦荟苦素的工艺研究

摘  要:研究比较了六种大孔吸附树脂HZ-801、HZ-806、HZ-818、HZ-841、HZ-830、HZ-816对芦荟苦素的吸附和解吸性能,从中筛选出适合芦荟苦素分离纯化的最佳树脂即HZ-830,并对该树脂的静态和动态吸附条件进行了研究,确定了树脂纯化芦荟苦素的最佳工艺条件,即:在25℃下,最佳上样浓度为0.5321mg/mL左右,吸附流速2BV/h,上样量为10BV,上样后分别用6BV的水和4BV的5%的乙醇洗脱,洗脱流速为3BV/h,经树脂一次柱纯化后芦荟苦素的纯度由9.58%提高到56.01%,回收率为75.9%,经过同一柱纯化体系进行二次上柱纯化后芦荟苦素的纯度由56.01%提高到85.8%,回收率为66.5%,二次纯化产品再经无水乙醇结晶的产物纯度可达95%以上。The performance and separation characteristics of six macroporous resin for purification of aloesin had been evaluated, including HZ-801, HZ-806, HZ-818, HZ-841, HZ-830, HZ-816. The result indicated that HZ-830 offered higher adsorption and desorption capacities. Then static and dynamic adsorption test was performd on the HZ-830 to optimize the purification process of aloesin from aloe. The optimum conditions were as follow,the volume,concentration,flow rate of loading sample were 10 bed volume(BV),0.5321mg/mL,2BV/h, respectively,at 25℃ desorption was performed successively with 6BV deionized water and 4BV 5% ethanol at eluting velocity of 3BV/h. After one run treatment,the content of aloesin in the product increased from 9.58% to 56.01% with a recovery of 75.9% ,then after a second treatment with the same process,the content was increased from 56.01% to 85.8% with a recovery of 66.5%. Finally,a high purity of aloesin product was obtained (purity ≥95%) after treating with crystallization.
