摘  要:目的研究骨髓间充质干细胞(BMSCs)与骨髓瘤细胞株共培养对成纤维细胞激活蛋白(FAP)、增殖诱导配体(APRIL)和B细胞激活因子(BAFF)表达的影响。方法体外分离、培养BMSCs,并鉴定其成骨分化能力。实验分为BMSCs单独培养的阴性对照组(A组)、BMSCs与U266细胞共培养组(B组)和BMSCs与RPMI8226细胞共培养组(C组)。培养7d和12d后,采用RT-PCR方法检测BMSCs中FAP、APRIL和BAFF mRNA表达水平的变化。结果三组BMSCs形态相似,均具有成骨分化能力。与A组相比,B、C组培养7d后,APRIL、FAP mRNA表达降低(P<0.05),而7d和12d后的BAFF mRNA表达无统计学差异(P>0.05)。结论 BMSCs与骨髓瘤细胞株U266或RPMI8226共培养后,改变FAP和APRIL表达,从而参与骨髓瘤的病理过程。Objective To investigate the effects of bone marrow mesenchymal stem cells (BMSCs) co-cultured with myeloma cell lines on the expressions of fibroblast active protein(FAP), a proliferation-inducing ligand (APRIL) and B-cell activating factor (BAFF) in BMSCs. Methods BMSCs were separated and cultured in vitro,and then the capability of osteogenic differentiation was identified. The BMSCs were divided into three groups of A (BMSCs alone as negative control), B (BMSCs co-cultured with U266 cells) and C(BMSCs co-cultured with RPMI8226 cells). The mRNA expressions of FAP,APRIL and BAFF were detected by RT-PCR after 7 and 12 days. Results The appearance of BMSCs of three groups was similar, and BMSCs all had the capability of osteogenic differentiation. Compared with group A, the mRNA expressions of APRIL and FAP after 7 days were decreased(P〈0. 05), while the mRNA expression of BAFF after 7 and 12 days was not significantly changed in groups of B and C(P〈0. 05). Conclusion The BMSCs co-cultured with myeloma cell lines U266 and RPMI8226 can down-regulate the expressions of APRIL and FAP,which might take part in the pathological process of multiple myeloma.
