摘 要:目的探讨丹酚酸B盐(SA-B)对转化生长因子β1(TGF-β1)刺激的肾间质成纤维细胞活化的作用及可能机制。方法 TGF-β1 2ng/ml刺激肾间质成纤维细胞NRK-49F后,分为对照组(C组)和SA-B 0.5、1、2、4、8μmol/L组(A1、A2、A3、A4、A5组),分别孵育24、48h,采用Westernblot检测Notch1、Jagged1、纤维连接蛋白(FN)的表达。结果与C组相比,不同浓度SA-B均能降低Notch1、Jagged1、FN蛋白表达;其中,A5组抑制效应最为明显(P<0.01),24h时Notch1、Jagged1、FN蛋白表达分别下降35.62%、34.39%、33.46%,48h时分别下降27.86%、30.79%、29.83%。结论 SA-B在体外可能通过下调Jagged1/Notch1信号通路,抑制TGF-β1诱导的成纤维细胞FN增加,从而防治肾间质纤维化。Objective To investigate the effect and underlying mechanism of salvianolic acid B (SA-B) on the activation of transforming growth factor-β1(TGF-β1)-stimulated renal interstitial fibroblasts. Methods The renal interstitial fibroblasts NRK-49F was stimulated by TGF-β1 2 ng/ml and then divided into 6 groups of C(normal controls), Al(treated with SA-B 0.5/~mol/L), A2(treated with SA-B 1 μmol/L), A3 (treated with SA-B 2 μmol/L), A4 (treated with SA-B 4 mol/L) and A5 (treated with SA-B 8 tlmol/L). At 24 and 48 h of culture, the protein expressions of Notch1, Jaggedl and fibronectin(FN) were detected by Western blot, respectively. Results Compared with group C, different concentrations of SA-B could inhibit the protein expressions of Notch1, Jagged1 and FN, especially in group A5 (P〈0. 01). The protein expressions of Notchl, Jagged1 and FN in group A5 were remarkably decreased by 35.62%, 34.39% and 33.46% at 24 h and 27.86%, 30.79%and 29.83% at 48 h, respectively. Conclusion SA-B inhibits TGF-β1-induced FN increase in the fibroblasts probably via down-regulating Jaggedl/Notchl signaling pathway, which may prevent renal interstitial fibrosis.
