摘 要:目的:研究肝癌组织与癌旁组织的基因表达谱差异。方法:应用包含44 000条人类全长基因的寡聚核苷酸芯片检测肝癌组织与癌旁组织的基因表达谱,并比较两者之间存在明显上、下调的表达差异基因,随后用RT-PCR和Western blot方法进行验证分析。结果:基因芯片筛选出存在差异表达的基因,其中沉默信息调节因子1(Sirt1)在肝癌组织与癌旁组织中的表达差异明显,在肝癌组织中明显升高,经RT-PCR和Wes tern blot证实与基因芯片结果一致。结论:Sirt1基因的表达可能与肝癌的发生发展密切相关。Objective: To explore the changes in gene expressions in tissues of patients with Hepatocellular carcinoma (HCC) by cDNA microarray. Method: A 44,000 gene cDNA microarray (Affymetrix) was used to examine the differential gene expressions between the tissues of patients with Hepatocellular carcinoma(HCC) and the tissues subjected to Real-time PCR analysis. Result: The of para-HCC. The Sirtl gene was identified and expression of Sirtl in tissues of HCC was extremely higher than in tissues of para-HCC. The results of Real-time PCR were consistent with those of genechip analysis. Conclusion: The expression of Sirtl was obviously differential between tissues of HCC and para-HCC. Sirtl may be associated with carcinogenesis of HCC.
