摘  要:目的:检测富含半胱氨酸61(Cyr61)蛋白、乙酰肝素酶(HPA-1)蛋白及Cyr61基因、HPA-1基因在结肠癌组织中的表达水平,探讨二者在结肠癌中侵袭转移的机制及关系。方法:用western blot方法检测结肠癌组织Cyr61和HPA-1的蛋白表达水平,用实时荧光定量逆转录聚合酶链反应(RT-PCR)方法检测结肠癌Cyr61和HPA-1的基因表达水平,分析二者与结肠癌的临床病理特征的关系。结果:结肠癌中Cyr61蛋白及HPA-1蛋白的表达水平高于癌旁组织和正常组织,癌旁组织高于正常组织(P均<0.001);结肠癌中Cyr61 mRNA及HPA-1 mRNA的表达水平高于癌旁组织和正常组织,癌旁组织高于正常组织(P均<0.001)。Cyr61蛋白及基因和HPA-1蛋白及基因表达的增强与肿瘤的分化程度、淋巴转移及TNM分期等相关(P均<0.05)。结论:Cyr61蛋白、HPA-1蛋白及Cyr61 mRNA、HPA-1 mRNA在结肠癌组织中呈高表达,可促进结肠癌的侵袭转移。Objective: The expression of Cyr61 protein HPA-1 protein and Cyr61 mRNA HPA-1 mRNA in colorectal neoplasm were detected, and also their correlation with invasion and metastasis was investigated in this research, Methods: The methods of western blot and real-time fluorescent quantitative (RT-PCR)were used to detect the expression of Cyr61 and HPA-lin specimens from 49 patients with colorectal cancer. Results: And the results of western blot were as follows: The expression of Cyr61 protein and Cyr61 mRNA were higher in colorectal neoplasms than that in the paired adjacent colorectal neoplasms and normal colorectal tissue(all P〈 0.001). The expression of HPA-1 protein and HPA-1 mRNA were higher in colorectal cancer than that in the paired adjacent colorectal neoplasms and normal colorectal tissue (all P〈0.001). Conclusion: The expression of Cyr61 and HPA-1 were significantly highest in colorectal cancer. At the same time, the increased expression of Cyr61 and HPA-1 can promote the invasion and metastasis of colorectal cancer.
