摘 要:目的:探讨胆固醇调节元件结合蛋白质1(SREBP1)在子宫内膜癌中的表达情况及SREBP1 shRNA对子宫内膜癌细胞AN3-CA生物学行为的影响。方法:采用免疫组织化学染色、实时定量逆转录聚合酶链反应、蛋白免疫印迹、Transwell体外细胞侵袭试验等方法分析SREBP1在子宫内膜组织中的表达及其对细胞生物学行为的影响。结果:SREBP1在正常子宫内膜组织中低表达,在子宫内膜癌组织中高表达,且免疫组织化学染色H-Score评分在子宫内膜癌高分化组和中、低分化组之间具有统计学差异(P<0.001);SREBP1 shRNA可减慢子宫内膜癌细胞AN3-CA的增殖速度并降低AN3-CA细胞的侵袭能力,均具有统计学差异(P<0.001)。结论:SREBP1的表达与活化参与子宫内膜癌的进展;SREBP1 shRNA可降低子宫内膜癌细胞的增殖和侵袭能力,表明内源性SREBP1参与子宫内膜癌的进展与转移,为子宫内膜癌潜在的分子靶向治疗提供了一定的理论基础。Objective:To detect sterol regulatory element binding protein1(SREBP1) expression in endometrial cancer and to explore the influence of SREBP1 shRNA on the biological behavior of endometrial cancer cells.Methods:Immunohistochemistry staining,quantitative real-time reverse transcription-polymerase chain reaction,Western blot,Transwell invasion assay were conducted to detect SREBP1 expression and to analyze the influence of SREBP1 shRNA on the biological behavior of endometrial cancer cells.Results:SREBP1 was expressed in normal endometrium and overexpressed in endometrial cancer;IHC H-Score ratings between highly differentiated endometrial cancer group and moderately to low differentiated group exhibited a significant difference(P0.001).SREBP1 shRNA decreased cell proliferation and invasion capacity of AN3-CA cells(P0.001).Conclusions:There was a close correlation between SREBP1 overexpression,over-activation and progression of endometrial cancer.SREBP1 shRNA reduces cell proliferation and invasion capacity,indicating that endogenous SREBP1 plays a role in development and metastasis of endometrial cancer.Furthermore,the results of this study indicate that SREBP1 could serve as a potential molecular target in the treatment of endometrial cancer.
