摘  要:目的研究N-糖基化抑制对胶质瘤细胞生长状态的影响。方法以依霉素(tunicamycin)为N-糖基化抑制剂,作用于人脑胶质瘤细胞株SWO-38,运用电镜技术及琼脂糖电泳,观察N-糖基化抑制对胶质瘤细胞生长的影响。结果依霉素作用于SWO-38细胞后,透射电镜下细胞出现线粒体等细胞器增殖,核仁消失,细胞团缩。琼脂糖电泳出现DNA梯形条带。结论N-糖基化抑制可以诱导胶质瘤细胞的早期凋亡。Objective The effect of N-glycosylation inhibition on the growth of glioma cells was observed in vitro. Methods Tunicamycin was used as Nglycosylation inhibitor and its effect on the glioma cell line SWO-38 was observed by transmission electron microscopy and DNA electrophoresis, Results The morphological changes characteristic of early apoptosis have been found, including proliferation of the cell organelle, such as mitochondria, reduction of cell volume, chromatin condensation. DNA ladder was also observed in tunicamycin treated cells. Conclusion N-glycosylation inhibition can induce early apoptosis in glioma cells.
