摘  要:目的探讨重组人促红细胞生成素(rhEPO)对高浓度葡萄糖作用下体外混合培养大鼠视网膜神经细胞凋亡的保护作用及对Bcl-2蛋白表达的影响。方法将体外混合培养3d的原代大鼠视网膜神经细胞随机分为高浓度葡萄糖培养组、对照组和各浓度rhEPO处理组,rhEPO处理组在高浓度葡萄糖培养液中分别加入终浓度为5、10、20、40kU/L rhEPO,培养48h后,采用MTT法检测视网膜神经细胞活力,免疫组织化学方法检测不同浓度rhEPO处理后高浓度葡萄糖损伤神经细胞中Bcl-2蛋白的表达水平。结果高浓度葡萄糖培养组细胞活力较对照组显著下降(F=74.54,q=21.85,P<0.01),但rhEPO处理组细胞活力均显著高于高浓度葡萄糖培养组(q=3.66~18.77,P<0.05、0.01)。高浓度葡萄糖培养组Bcl-2蛋白的表达较对照组明显下降(F=53.84,q=3.56,P<0.05),各rhEPO处理组Bcl-2蛋白的表达明显高于高浓度葡萄糖培养组(q=10.13~19.14,P<0.01)。结论rhEPO能减少体外培养的高浓度葡萄糖培养状态下视网膜神经细胞的凋亡,其机制可能与上调Bcl-2的表达有关。Objective To investigate the protection of recombinant human erythropoietin (rhEPO) on apoptosis of retinal nerve cells cultured in mixed condition with high concentration of glucose in vitro and the expression of Bcl-2. Methods The pri- mary retinal cells of rat cultured for three days were randomized to high-concentration glucose (HCG) group, control group, and treatment groups with different concentrations of rhEPO. Different doses of rhEPO (5, 10, 20 and 40 kU/L) were addeded into the treatment groups and cultured for 48 h. MTT assay was used to estimate the vitality of the cells, and the expressions of Bcl-2 cells were detected by semi quantitative immunocytochemistry. Results The cytoactive cultured in HCG was markedly lower than that in the control group (F=74.54,q=21.85,P〈0.01), but that after treatment with rhEPO was higher than that in HCG group (q= 3.66-18.77;P〈0.05,0.01). The expression of Bcl-2 protein in HCG group was lower than that in the control (F = 53.84,q=3.56,P〈0.05), but that after treatment with different concentrations of rhEPO was higher than that in HCG group (q= 10.13-19.14,P〈0.01). Conclusion rhEPO can decrease the apoptosis of retinal nerve cells cultured in vitro under HCG environment, which is probably associated with up regulation of Bcl-2 expression.
