摘　　要：目的研究结肠癌组织中MTA1的表达水平及在细胞核、细胞质内的定位情况,分析其与结肠癌预后的相关性,并探讨其可能的机制。方法采用免疫组织化学染色方法在结肠癌组织芯片(30例肠癌组织、10例转移组织、8例正常组织)中检测MTA1与Sox2蛋白的表达水平及定位情况,统计分析二者与临床病理资料及预后的相关性;并初步探讨Sox2与MTA1表达的相关性。结果①肠癌组织及转移组织中MTA1细胞质染色强度均显著高于正常组织(P<0.05),而肠癌组织与转移组织间MTA1表达差异不显著(P>0.05);②MTA1在细胞质的表达强度与临床分期呈正相关(P=0.03),生存时间<5年的肠癌患者的肿瘤组织细胞质内MTA1的表达强度显著高于生存时间≥5年的患者组织(P=0.007);③Sox2在肠癌组织细胞核及细胞质内的表达均与MTA1细胞质表达具有显著的相关性(P<0.01)。结论 MTA1细胞质过表达提示结肠癌预后不良,这可能与Sox2的过表达相关。Objective To assess the expression and distribution of MTA1 in colon cancer and explore the correlation between MTA1 and prognosis of colon cancer and its mechanism. Methods Immunohistochemistry staining was used to e- valuate the expression of MTA1 and Sox2 proteins in colon cancer tissue array, which included 30 cases of cancer tissues, 10 cases of metastases and 8 cases of normal tissues. The correlation between MTA1 expression and the clinical and patho- logical parameters, as well as that between MTA1 and Sox2, was analyzed. Results （1） MTA1 expression in tumor tis- sues and metastases was significantly stronger than that in normal tissues （ P 〈 0. 05 ）, but no significant difference was found between primary and metastatic cancer tissues; （2） The expression of MTA1 in cytoplasm was positively correlated with the clinical stage of the cancer （ P 〈 0.05 ）. The cytoplasm expression of MTA1 was significantly stronger in colon cancer patients with survival time 〈 5 years than that of patients with survival time ≥5 years （P 〈0. 05） ; （3） Significant correlation was observed between cytoplasm expression of MTA1 and nucleus or cytoplasm levels of Sox2 （ P 〈 0. 01 ）. Conclusion The over-expression of MTA1 in cytoplasm positively correlates with the poor prognosis of colon cancer pa- tients, which might be related to the over-expression of Sox2.
