摘 要:目的 明确妊娠滋养细胞疾病和正常绒毛组织中的端粒酶表达状况 ,探索端粒酶活化在滋养细胞肿瘤形成中的作用。方法 应用以聚合酶链反应为基础的端粒酶测定方法 (TRAP) ,对不同临床类型妊娠滋养细胞疾病及不同发育阶段正常妊娠绒毛组织共 61例的端粒酶活性进行检测。结果 绒毛膜癌及侵蚀性葡萄胎 36.4% (4/1 1 )端粒酶阳性 ,葡萄胎 2 6.3% (5/1 9)阳性 ;早孕 60 d以内绒毛全部 (1 0 /1 0 )阳性 ,此期以后的胎盘组织 (2 1 /2 1 )阴性。结论 端粒酶在妊娠早期以后表达受到抑制 ,其活化在滋养细胞肿瘤形成中可能起重要作用 ;其测定对此类疾病的诊断。Objective To investigate the prevalence of telomerase expression in gestational trophoblastic diseases and chorionic villi, and to evaluate the role of telomerase activation in tumorigenesis of tropholbastic tumors.Methods The telomeric repeat amplification protocol (TRAP) assay was used to detect telomerase activity in 61 samples from a variety of gestational tropholbastic tumors and different developing periods of normal chorionic villi.Results Telomerase activity was found in 36.4%(4/11) of gestational trophoblastic tumors (including choriocarcinomas and invasive moles),26.3%(5/19) of hydatidiform moles and 100%(10/10) of early embryonic villi, but was negative in the 21 cases of normal chorionic villi after 60 d of pregnancy .Conclusion Telomerase is expressed in the villi during very early embryonic period, but is repressed after the completion of organogenesis.Telomerase activation may play an important role in the tumorigenesis of trophoblastic tumors.Detection of telomerase activity has some potential value in differential diagnosis, prognosis and chemotherapeutic effect evaluation of these diseases.
