摘 要:目的检测肝癌热消融治疗后调节性T细胞(Tregs)的变化,探讨其免疫增强的相关机制。方法流式细胞仪检测Tregs细胞的比例,荧光定量PCR检测Foxp3基因的表达。结果对照组、治疗组消融治疗前和治疗后Tregs比例分别为(3.66±0.42)%、(11.75±1.21)%、(5.74±0.65)%(F=12.64,P<0.01)。各组中淋巴细胞Foxp3的表达量分别为(0.331±0.051)、(0.762±0.104)、(0.446±0.052)(F=11.37,P<0.01)。结论下调Foxp3表达抑制CD4+T细胞向Tregs分化可能是热消融免疫增强作用的主要作用机制。Objective To investigate the changes of Treg cells in liver cancer patients with thermal ablation treatment. Methods The percents of Treg cells was detected by flow cytometry analysis. The mRNA expression of Foxp3 was deter mined by Real-time PCR analysis. Results The percent of Treg cells was(3.66±0.42)%, (11.75±1.21)%,and(5.74± 0.65) % in contol group, before ablation treatment group, and after ablation treatment group, respectively (F = 12.64, P 〈 0.01 ). The mRNA expression of Foxp3 in those groups was(0.331±0.051 ), (0.762±0.104),and (0.446±0.052),respectively (F = 1.1.37 ,P 〈 0.01 ). Conclusion Thermal ablation treatment enhanced immunity in in hepatocellular carcinoma patients, and which was completed by downregulation of Foxp3 gene to inhibit the diffination of CD4+cells toTreg ceils.
