摘　　要：目的本研究拟通过荟萃分析评价脂联素基因(ADIPOQ)三个多态位点,包括rs2241766、rs1501299和rs266729与脑卒中和高血压的相关性。方法通过文献检索,收集国内外相关研究文献,应用固定效应模型和随机效应模型两种方法合并比值比(OR)及相应的95%置信区间(95%CI),同时检验研究间的异质性。结果本项荟萃分析共纳入16项研究,其中脑卒中荟萃分析纳入1,586个病例和5,426个对照;高血压荟萃分析纳入1,751个病例和3,025个对照。rs2241766和rs266729与脑卒中存在显著关联。固定效应模型合并OR值(95%CI;P值)分别为1.28(95%CI:1.12,1.46;P<0.0001)和1.20(95%CI:1.08,1.34;P=0.0001)。在rs2241766与脑卒中的荟萃分析中发现纳入的研究间存在显著异质性(I2=70%,P=0.01),应用随机效应模型合并OR值为1.26(95%CI:0.98,1.63;P=0.07)。三个位点与高血压之间关联都没有统计学意义。结论本研究结果提示ADIPOQ基因单核苷酸多态性rs2241766和rs266729与脑卒中之间存在显著关联。Objective The aim of this study was to evaluate the relationship between three single nucleotide polymorphisms（SNPs）,rs2241766,rs1501299 and rs266729 in adiponectin gene（ADIPOQ） and stroke,as well as hypertension.Methods A comprehensive search was conducted to identify all studies on the association of ADIPOQ gene polymorphisms with stroke and hypertension.The fixed or random effects model was selected to calculate the pooled odds ratios（ORs） and corresponding 95% confidence intervals（95%CIs） based on the homogeneity test among studies.Heterogeneity among studies was evaluated using Q test and the I2.Results A total of 1,586 cases and 5,426 controls for stroke and 1,751 cases and 3,025 controls for hypertension were enrolled in this meta-analysis.The SNPs rs2241766 and rs266729 were significantly associated with stroke,yielding pooled ORs（fixed-effects method） of 1.28（95%CI：1.12,1.46;P0.0001） and 1.20（95%CI：1.08,1.34;P=0.0001）,respectively.Significant between-study heterogeneity was found for rs2241766 in our meta-analysis of stroke（I2=70%,P=0.01）.The random-effects pooled OR for rs2241766 was 1.26（95%CI：0.98,1.63;P=0.07）.The associations between the three SNPs and hypertension were not significant.Conclusion The present meta-analysis showed that the associations between rs2241766 and rs266729 in the ADIPOQ and stroke were significant.None of the three SNPs were significantly associated with hypertension.
