摘  要:目的:通过检测信号转导及转录活化因子3(STAT3)在非小细胞肺癌(NSCLC)患者外周血中的表达水平,探讨STAT3作为潜在肿瘤标志物对NSCLC的诊断价值。方法采用RealtimePCR检测STAT3 mRNA、ELISA方法检测STAT3蛋白在NSCLC患者、肺良性疾病患者、正常人外周血中的表达;应用ROC曲线评价STAT3的诊断价值。结果STAT3 mRNA、STAT3蛋白在肺癌组的表达水平高于肺良性疾病组及正常对照组(P<0.05或P<0.01);STAT3 mRNA和STAT3蛋白的ROC曲线下面积(Az)分别为0.870、0.860,两者的Az比较,差异无统计学意义(P=0.500)。结论STAT3在NSCLC患者外周血中呈异常高表达,其对NSCLC具有中等的诊断价值,有可能成为一种新的NSCLC相关标忠物。Objective To explore the clinical diagnostic value of signal transducer and activator of transcription 3 (STAT3) as a potential tumor marker by detecting the expression of STAT3 in blood of non-small cell lung cancer(NSCLC) patients. Methods Real time PCR and ELISA was separately applied to detect the expression of STAT3 mRNA and STAT3 protein in the NSCLC patients,benign pulmonary disease patients and normal people. The ROC curve was used to analyze the diagnostic value of STAT3 in NSCLC. Results The expression levels of STAT3 mRNA and STAT3 protein in lung cancer group were significantly higher than those in benign pulmonary disease group and normal control group ( P 〈d0.05 or P〈 d0.01). The area under ROC curves (Az) of STAT3 mRNA and STAT3 protein were 0. 870,0. 860 respectively,the Az between two groups had no significant difference ( P =0. 500). Conclusions STAT3 is overexpressed in blood of NSCLC patients. STAT3 has medium diagnostic value and may be a new tumor marker of NSCLC.
