摘　　要：目的探讨125I籽粒近距离照射对胶质瘤干细胞的生长抑制作用。方法体外分离培养人胶质瘤干细胞,按照射总剂量分为对照组、10 Gy组、20 Gy组和30 Gy组,应用125I籽粒近距离照射胶质瘤干细胞,台盼蓝染色鉴定照射后细胞死亡率,克隆形成实验鉴定照射后细胞增殖能力。结果细胞死亡率:对照组(7.05±0.21)%,10 Gy组(47.92±0.35)%,20 Gy组(60.77±0.41)%,30 Gy组(84.32±0.53)%;与对照组比较,10~30 Gy组细胞死亡率差异均有统计学意义(P<0.05)。细胞生长分数:对照组为1,10 Gy组为0.75±0.06,20 Gy组0.52±0.04,30 Gy组0.25±0.02;与对照组比较,10~30 Gy组细胞生长分数差异均有统计学意义(P<0.05);且生长分数与照射剂量呈负相关(γ=-0.981,P<0.05)。结论125I籽粒近距离照射可抑制胶质瘤干细胞的增殖并导致细胞死亡。Objective To explore the inhibitory effect of 125I radiation at a short distance on glioma stem cells.Methods Glioma stem cells were isolated,cultured in vitro and randomly divided into control group,10 Gy group,20 Gy group and 30 Gy group according to irradiation doses.The glioma stem cells were irradiated in a short distance by 125I seeds.After irradiation,trypan blue staining was carried out to identify the cell mortality and colony formation assay was adopted to detect the cell proliferation.Results The cell mortality was 7.05±0.21 % in control group,47.92±0.35 % in 10 Gy group,60.77±0.41 % in 20 Gy group and 84.32±0.53 % in 30 Gy group,and there was a statistical significance in the differences between the 10 to 30 Gy group and the control group(P<0.05).The cell growth fraction was 1 in control group,0.75±0.06 in 10 Gy group,0.52±0.04 in 20 Gy group and 0.25±0.02 in 30 Gy group,and there was a statistical significance in the differences between the 10 to 30 Gy group and the control group(P<0.05),and there was negative correlation between the cell growth fraction and irradiation doses(γ=-0.981,P<0.05).Conclusion 125I irradiation in short distance can inhibit the cell proliferation and lead to cell death in glioma stem cells.
