摘  要:目的 为脑膜瘤X 刀的个体化治疗寻找最佳量 效关系和最佳时 效关系。方法  8例病人的脑膜瘤细胞进行原代培养。将培养的脑膜瘤细胞分为阴性对照组、5Gy、10Gy、15Gy、2 0Gy、2 5Gy组和 30Gy组 ,培养 7~ 9天后采用单次X线照射 ,于照射后 2h、6h、12h、2 4h、48h和 72h采用FCM检测细胞凋亡和细胞存活情况 ,采用免疫组化(ABC)法测试PCNA、BcL 2和p5 3基因的表达。结果 发现原代培养的脑膜瘤细胞于培养后的第 7~ 2 1天 ,细胞存活率可达 80 %~ 92 %。X线诱导细胞凋亡平均最佳剂量为 2 0Gy,平均最佳检测时间为 2 4h ,平均凋亡率为 5 7 6 5 %± 15 5 2 %。结论 不同的个体需要的细胞凋亡的X线剂量有很大差异。尽管影响剂量选择的因素很多 。Objective To find the best relationship of dose effect and time effect for X knife treatment meningioma.Methods Meningioma cells from 8 patients were cultured.Cells from 5 cases were used in experiment and the left were used to do preliminary test.In each case,cells were divided into 7 groups:5Gy,10Gy,15Gy,20Gy,25Gy,30Gy and negative control.After cultured for 7~9days,Varian Clinac 600C was used to radiate meningioma cells.After 2h,6h,12h,48h,72h and 96h,cell apoptosis was detected by Flow Cytometer (FCM) and morphological check.Immunohistochemistry(ABC) method was used to examine the expression of p53,BcL 2 and PCNA genes.Results Cell survival rate was in the range of 80%~92% during the 7th 21st day.The largest apoptotic rate was achieved 24h after 20Gy radiation.The average apoptotic rate was 57.65%±15.52%.Conclusion The dosage needed to induce cell apoptosis of these individuals are significantly different.Many factors are shown to be involved in it.Our experiment indicate that p53,BcL 2 and PCNA are one of these important factors.
