摘  要:目的观察L型氨基酸转运载体1(LAT1)与其异二聚体CD98hc在胃癌细胞SGC-7901中的表达及LAT1对细胞增殖与迁移能力的影响。方法将胃癌细胞SGC-7901分为1、10、100μmol/L BCH干预和空白对照共4组,RT-PCR、IF与Western blot检测胃癌细胞SGC-7901中LAT1与CD98hc mRNA、蛋白的表达。不同浓度的BCH作用于胃癌细胞SGC-7901 48 h后,新型四唑氮盐(MTS)比色法检测各组细胞生长情况,流式细胞仪检测各组细胞周期分布情况,Transwell细胞迁移小室检测各组细胞迁移能力的变化。结果 LAT1及其异二聚体CD98hc在胃癌细胞SGC-7901中呈高表达。与空白对照组相比,随着BCH浓度的提高,胃癌细胞SGC-7901的增殖能力与迁移能力逐渐降低。细胞周期分析发现,BCH处理的胃癌细胞组S期细胞减少,而G0/G1期细胞增加(P<0.05)。结论 LAT1及其异二聚体CD98hc在SGC-7901细胞中有表达,LAT1可能促进SGC-7901细胞的增殖,加快细胞周期进程,增强SGC-7901细胞的迁移能力。Objective To observe the expression of L amino acid transporter 1 ( LAT1 ) and its heterodimer CD98hc in gastric cancer SGC-7901 cells and the effect of LAT1 on proliferation and migration of SGC-7901 cells. Methods Gastric cancer SGC-7901 cells were divided into 1 μmol/L BCH intervention group, 10 mol/L BCH intervention group, 100 μmol/L BCH intervention group and control group. Expression of LAT1 and CD98hc mRNA in gastric cancer SGC-7901 cells was detected by RT-PCR, IF and Western blotting, respectively. After the SGC-7901 cells were cultured with BCH at different concentrations for 48 h, their prolif- eration was detected by MTS colorimetry, distribution of cell cycle in different groups was assayed by flow cy- tometry, and cell migration in different groups was observed in Transwell chamber. Results LAT1 and CD98hc were highly expressed in SGC-7901 cells. The proliferation and migration level in SGC-7901 cells de- creased with the increasing BCH concentration and were significantly lower in 3 BCH intervention groups than in control group. Cell cycle analysis showed that the number of SGC-7901 cells was lesser in S phase and greater in G0/Gj phase after BCH treatment(P 〈 0. 05 ). Conclusion LAT1 and CD98hc are expressed in SGC-7901 cells. LAT1 may promote the proliferation and migration of SGC-7901 cells and speed up their cell cycle process.
