人卵巢癌顺铂耐药细胞EZH2基因沉默对Drosha基因表达的影响

摘  要:目的探讨RNA干扰沉默果蝇zeste基因增强子同源物2(EZH2)基因对卵巢癌获得性耐药细胞株A2780/DDP的Drosha基因表达的影响。方法获得稳定表达EZH2短发夹状RNA(shRNA)的人卵巢癌获得性耐药细胞株A2780/DDP-shEZH2,实时荧光定量PCR法和免疫印记法检测转染株的Drosha基因表达。结果以未转染组A2780/DDP细胞Drosha的表达水平为100.00%,EZH2-shRNA转染组A2780/DDP-shEZH2细胞Drosha的mRNA及蛋白水平分别上升(53.33±9.24)%和(51.28±3.25)%。结论沉默EZH2基因可以上调Drosha基因的表达。Objective To investigate the effect of gene silencing of RNA interfering enhancer of zeste homolog 2 (EZH2) on Drosha gene expression in cisplatin-resistant ovarian cancer cell line A2780/DDP. Methods After the silencing EZH2 A2780/DDP cell line was obtained by short hairpin RNA(shRNA) technique,the mRNA and protein expression of Drosha were detected by real-time qRT-PCR and Western blot. Results The Drosa expression of A2780/DDP in untransfection group was 100%, and the expressions of Drosha mRNA and protein in EZH2-shRNA transfection group increased by (53.33±9.24 )% and (51.28±3.25 )%, respectively. Conclusion Silencing EZH2 can increase the expression of Drosha gene.
