摘  要:目的探讨卒中后抑郁(post-stroke depression,PSD)模型大鼠杏仁核神经元PTEN/Akt信号通路的改变。方法成年健康雄性SD大鼠40只,随机分为对照组、模型1d组、模型7d组、模型14d组和模型21d组。采用颈总动脉线栓再灌注法制备局灶性脑缺血大鼠模型,结合孤养和慢性不可预见的温和性应激方法制备PSD大鼠模型。采用流式细胞术检测杏仁核神经元凋亡率,免疫印迹方法检测PTEN和p-Akt的表达。结果 (1)模型各组杏仁核神经细胞凋亡率明显高于对照组(P<0.05);(2)各模型组杏仁核组织PTEN表达均高于对照组,p-Akt表达低于对照组,差异均有统计学意义(P<0.05)。结论 PSD模型大鼠杏仁核存在明显的神经元凋亡,这可能与PTEN上调和Akt下调有关。Objective To observe the change of PTEN/Akt signal pathway in amygdala of post-stroke depression (PSD) model rats. Methods Forty male SD rats were randomly divided into control group and model groups (on ld,7d, 14d and 28d). The rat model of post-stroke depression was set up by thread occlusion through arteria carotis interna and reperfusion. Then the rats were raised alone and given chronic unpredictable modest stress to establish the PSD model. Double-labeled flow cytometry (FCM) was employed for the quantification of the apoptotic cells in the amygdala, and the expression of PTEN and p-Akt was detected by Western-blotting. Results ( 1 ) The apoptotie cell rate of every model group was higher than that of control group ( P 〈 0.05 ). ( 2 ) Campared to control rats, the PTEN expression of model rats was higher,while the p-Akt was lower ( P 〈 0.05 ). Conclusion Apoptosis was increased in amygdala of PSD model rats, and might be associated with the change of PTEN/Akt signal pathway.
