摘  要:目的应用D-半乳糖(D-galactose,D-Gal)制备亚急性衰老动物模型并对该模型进行评价和探讨。方法将16只SD大鼠随机分为两组,腹腔注射D-半乳糖建立亚急性衰老动物模型,采用Y型迷宫检测行为学变化;以TUNEL染色观察海马CA1区神经元凋亡;以免疫组化检测大鼠海马CA1区神经型一氧化氮合酶(neuronal nitric oxide synthase,nNOS)及半胱氨酸天冬氨酸蛋白酶9(cysteinyl aspartate-specific protease,caspase-9 P35)表达。结果与空白对照组相比,衰老模型组学习尝试次数明显增加(77±5.2)次及(50±6.3)次,记忆正确次数明显减少(3.7±0.8)次及(7.7±0.5)次,差异均有统计学意义(P<0.01);衰老模型组较空白对照组海马CA1区TUNEL阳性细胞(37.8±1.3)个/视野及(0.8±0.2)个/视野及caspase-9 P35表达(37.6±1.8)个/视野及(6.2±1.3)个/视野明显增加(P<0.01),而nNOS表达(6.8±1.5)个/视野及(22.8±3.6)个/视野明显降低(P<0.01)。结论应用D-半乳糖可成功制备亚急性衰老动物模型,体现了学习记忆障碍的行为学变化,又能较好地模拟老年性痴呆的特征。Objective The development and estimation of an animal subacute aging model induced by D-gal were reported in the paper. Methods 16 rats were randomly divided into two groups. The subaeute aging model were established by injecting intraperitoneally with the D-gal. Y-electrical maze test was used to determine the ability of learning memory. TUNEL method was used to detect apoptosis of hippocampal neurons in the CAl region. Immunohistochemistry were used to determine the expression of nNOS and caspase-9 P35 in hippocampus CA1 region. Results In the subacute aging model group, the learning times of Y-electrical maze were increased ( 77 ± 5.2 ) vs ( 50 ± 6.3 ) and the memory times were de- creased (3.7 ± 0.8 ) vs (7.7 ± 0.5 ) than the control group( P 〈 0.01 ). Compared with the control group, TUNEL-positive neurons (37.8±1.3) vs (0.8±0.2) and the activity of caspase-9 P35 (37.6 ± 1.8) vs (6.2 ± 1.3) had significant in- crease in hippocampal CA1 subfield of the subacute aging model group (P 〈 0. O1 ), while the activity of nNOS (6.8 ± 1.5) vs (22.8 ± 3.6) were conspicuously lower (P 〈 0. 01 ). Conclusion The subacute aging model can be established by in- tra-peritoneum injection of by D-gal, to reflect behavior variation caused by learning memory impediment, and to imitate the character of senile dementia better.
