摘  要:目的本实验主要建立运动病模型,观察运动病动物学习记忆能力的改变以及脑组织几种生化指标的变化,观察地芬尼多对运动病动物学习记忆能力的改善以及脑组织几种生化指标的变化,探讨地芬尼多对运动病产生治疗作用的机制。方法利用昆明种小鼠复制运动病模型,观察地芬尼多对运动病小鼠异食癖、学习记忆成绩以及游泳时间的影响;利用Wistar大鼠复制运动病模型,观察地芬尼多对运动病大鼠不同脑组织中生化指标的影响。结果模型组小鼠高岭土摄入量显著增加,表明实验性运动病小鼠模型复制成功。地芬尼多使运动病刺激后小鼠学习记忆成绩提高;降低运动病大鼠海马组织中一氧化氮、丙二醛含量,升高超氧化物歧化酶活力。结论地芬尼多可使脑血管紧张度降低,改善脑血流的分布,增加脑血流,致海马组织局部缺血缺氧得以缓解,氧自由基减少有关,调整脑细胞内NO、MDA、SOD等酶的活性,增强脑细胞的兴奋性,从而保持中枢神经系统的兴奋性,起到抗运动病的作用。Objective To study the effects of the experimental motionsickness on the performance of the mice, and the effect of Difenidol on them, and to study its probable mechanisms. Methods Kunming spices mice were taken to estab- lish motinsickness model and the the effect of Difenidol on pica,learning and memory scores of motinsickness mice was ob- served. Wistar white rats were taken to establish motinsickness model and the the effect of Difenidol on general conditions, pica, blood sugar, glycogen,lactic acid, lactate dehydrogenase and other biochemical index at different areas on motinsick- ness rats'brain were observed. Results It indicated that motionsickness model had been successfully eatablished. Difenidol could enhance the motionsickness mice'learning scores, ameliorate the pica of motionsickness mice, decrease the nitrous ox- ide and malondialchehyche contents in mice" hippocampi, and improve the superoxide dismutase in mice" hippocampi. Conclusions Difenidol can decrease the contraction of brain vessel, improve the distribution of blood flow, and increase blood flow which could relieve partial loss of blood supply and hypoxia in hippocampus and decrease the quantity of free radicals. It might also adjust NO, MDA and SOD in brain cell, increase the excitability of brain cell, then keep the excitabili- ty of the central nervous system which might resist motion sickness.
