摘 要:目的探讨雷公藤甲素促进人类鼻咽癌CNE-1细胞凋亡的机制。方法 MTT法检测雷公藤甲素对CNE-1的细胞毒作用;Hoechst 33258染色荧光显微镜观察细胞凋亡形态学改变;Western Blot检测细胞凋亡相关蛋白的变化。结果雷公藤甲素对CNE-1有强大的细胞毒作用,IC50为0.029±0.007μmoL/L;该化合物可浓度依赖性地诱导CNE-1细胞发生凋亡,出现典型的凋亡小体及细胞凋亡相关蛋白Caspase-3的裂解活化。结论雷公藤甲素可浓度依赖性地诱导人鼻咽癌CNE-1细胞发生凋亡,且与Caspase-3的裂解活化有关。Objective To investigate triptolide induced cell apoptosis in nasopharyngeal carcinoma (NPC) CNE-1cells. Methods The cytotoxicity of Triptolide to CNE-1 cells was measured by MTT assay; Apoptotic morphology was observed after Hoeehst 33258 staining; Changes of apoptotic related proteins were analyzed by western blot. Results Triptolide exhibited potent cytotoxicity against NPC CNE-1 Ceils, and IC50 was 0.029±0.007 μmoL.L-1. After CNE-1 cells were treated with different concentrations of triptolide for 24 h, typical apoptotic bodies and activation of Caspase-3 were detected in a dose-dependent manner. Conclusion Triptolide induced CNE-1 cancer cells apoptosis in a dose-dependent manner, which correlation with caspase-3 activation.
