初发和复发膀胱尿路上皮癌中miR-21和miR-205的表达

摘　　要：目的检测microRNA-21(miR-21)与microRNA-205(miR-205)在初、复发膀胱尿路上皮癌中的表达情况,探讨microRNAs(miRNAs)在膀胱尿路上皮癌中的表达意义及是否与肌层浸润有关。方法采用基于2-△△CT的实时定量PCR(Real-time PCR)方法检测40例初、复发癌组织miR-21及miR-205的表达情况。结果与初发癌相比,复发癌miR-21表达上调(P<0.05),miR-205表达下调(P<0.05);与非肌层浸润性癌相比,肌层浸润性癌miR-21表达上调(P<0.05),miR-205表达下调(P<0.05);肌层浸润性癌miR-21与miR-205表达量的比值5倍于其在非肌层浸润性癌中表达量的比值。结论 miR-21高表达、miR-205低表达可能参与膀胱尿路上皮癌复发并与肌层浸润有关。Objective To detect the expressions of miR -21 and miR -205 in maiden and recurrence bladder urothelial cell carcinoma and to explore the relationship between microlRNAs（miRNAs） and muscle -invasion in urothelial cell carcinoma of bladder. Methods 2- ΔCT Real -time PCR method was used for quantitative analysis of the expression of miR - 21 and miR - 205 in 40 patients with maiden and recurrence urothelial cell carcinoma of bladder. Results Compared with maiden tissues, the recurrence tissues showed significant miR - 21 over - expression （ P 〈 0.05 ） and miR - 205 down - regulation （ P 〈 0.05 ） ; Compared with non muscle - invasive bladder cancer, muscle - invasive bladder cancer showed significant miR - 21 over - expres- sion （ P 〈 0.05 ） and miR - 205 down - regulation（ P 〈 0.05 ） ; both maiden and recurrence cancer character- ized as muscle - invasive bladder showed a miR - 21 ： miR - 205 ratio at least 5 - fold higher than the quantita- tive ratio obtained from non muscle - invasive bladder. Conclusion The over expression of miR - 21 and lower expression of miR -205 may participate in recurrence urothelial cell carcinoma of bladder and have rela- tionship with muscle invasive.
