摘  要:目的:检测骨形态发生蛋白-2(BMP-2)基因mRNA及其蛋白的表达,探讨BMP-2基因修饰自体骨髓间充质干细胞(BMSCs)移植对兔下颌骨牵张成骨新骨形成的促进作用。方法:取新西兰白兔36只随机分为3组,每组12只。建立牵张成骨动物模型,在固定期第2天,实验组于牵张间隙注射200μL的BMP-2基因修饰的自体BMSCs液;对照组注射等量自体BMSCs液;空白组注射等量生理盐水。分别于固定2,6周通过逆转录多聚酶链式反应(RT-PCR)、免疫组化等手段检测BMP-2基因mRNA及其蛋白的表达情况。结果:实验组牵张间隙新生骨组织均可见BMP-2基因mRNA和其蛋白强阳性表达。结论:BMP-2基因修饰的自体BMSCs移植能有效促进兔下颌骨牵张成骨新骨形成。Objective:To examine the expressions of both bone morphogenetic protein-2 (BMP-2) gene mR- NA and related proteins and investigate the promotive effect of transplantation of BMP-2 gene transfected autogenous bone mesenchymal stem cells on bone regeneration in rabbit mandibular distraction osteogene- sis. Methods: Thirty-six New Zealand's white rabbits were randomly divided into three groups with twelve in each. All objects were prepared into distraction osteogenesis surgical model on right mandibles. On the 2^nd day of consolidation, experimental, control, and blank groups were injected with the same amount of 200 μL of the solution with BMP-2 gene transfected autogenous bone mesenchymal stem cells, the solution with autogenous bone mesenchymal stem cells, and physiological saline at distraction gap, respectively. The expressions of BMP-2 mRNA and related proteins were examined by RT-PCR and immunohistochem- istry at the ends of the 2nd and 6th week consolidations, respectively. Results: Strongly positive expression of both BMP-2 gene mRNA and related proteins were confirmed on regenerated bone in distraction gap. Conclusion: The transplantation of BMP-2 gene transfected autogenous bone mesenchymal stem cells could effectively promote bone regeneration in rabbit mandibular distraction osteogenesis.
