摘  要:目的观察大鼠海马结构在吗啡耐受中的作用。方法 SD大鼠20只随机分为两组:①对照组:背部皮下注射与耐受组等量的生理盐水;②耐受组:背部皮下一日三次逐日递增的注射盐酸吗啡,连续6天后,停止注射,处死全部大鼠,用免疫组化染色方法观察各组海马结构内的M1受体、环磷腺苷反应元件结合蛋白(cAMP response element binding protein,CREB)和c-fos蛋白表达。结果①在光镜下,HE染色显示吗啡耐受组与对照组相比未发现海马结构形态学有异常改变。②耐受组与对照组相比,组织化学染色发现M1受体表达减少,而c-fos和p-CREB的蛋白表达增加。结论海马结构可能参与了吗啡耐受,这可能与海马结构的M1受体表达减少有关,也与转录因子c-fos和p-CREB激活表达有关。Objective To detect the action of hippocampal formation in the morphine toler- ance. Methods Twenty SD rats were randomly divided into 2 groups:①control group: rats were subcutaneously injected the same amount of saline; ②tolerance group: morphine hydrochloride was administrated subcutaneously on the back three times a day and daily increments. Six days later, all the rats were killed, then changes of M1 receptor, p-CREB and c-los proteins expression were studied by using immunohistochemical method in hippocampal formation in both groups. Results ①No abnormal structure in hippocampal formation was found in tolerance group and control group under light microscope. ②Compared with control group, M1 receptor protein expression decreased and the expression of c-los protein and p-CREB protein increased in tolerance group. Conclusion Hippoeampal formation maybe involved in the morphine tolerance, which maybe related with M1 receptor protein expression reducing as well as the activated expression of transcription factors of c-los and p-CREB proteins.
