摘 要:目的研究光动力联合塞来昔布对子宫颈癌细胞中环氧合酶-2(cyclooxynase-2,COX-2)蛋白表达及细胞增殖和凋亡的影响。方法①培养子宫颈癌Hela细胞,应用四甲基偶氮唑蓝(MTT)比色法检测选择性COX-2抑制剂塞来昔布(celecoxib)对子宫颈癌Hela细胞增殖抑制作用。②将Hela细胞分为4组:空白对照组(Hela,H组)、塞来昔布组(celecoxib,C组)、光动力组(photodynamics therapy,PDT,P组)、塞来昔布+光动力组(celecoxib+photodynamics therapy,P+C组)、分别应用蛋白印迹法(Westernblot)和免疫组化法检测COX-2蛋白表达情况。结果 MTT比色法检测显示,celecoxib作用Hela细胞24 h后对宫颈癌细胞有相对的抑制作用。Western blot和免疫组化法检测结果均显示:celecoxib联合PDT作用Hela细胞24 h后COX-2蛋白的相对表达水平明显降低,与其它3组相比,差异有统计学意义(P<0.05)。结论选择性COX-2抑制剂celecoxib能抑制人子宫颈癌细胞的增殖,且呈浓度依赖性,随着浓度的增大作用逐渐增强。PDT联合选择性COX-2抑制剂celecobix能有效抑制靶基因COX-2的表达,进而可抑制子宫颈癌细胞增殖并诱导细胞调亡增加,为子宫颈癌的基因研究及治疗提供新思路。Objective To study the effects and expression of cyclooxynase-2 of photodynamics therapy combined with celecoxib on the proliferation and apoptosis in cervical carcinoma cells. Methods ① Hela cells were cultured, the proliferation and suppressive effects of using celecoxib in cervical cancer cell were detected by MTY. ②Hela cells were divided into control ( H), celecoxib ( C ) , PDT (P) and PDT + celecoxib ( P + C ) group. The cyclooxygenase-2 protein expression was detected by Western blot and immunohistochemistry technique. Results For 24 hours, cervical carcinoma cells was suppressed relatively by MTY. Photodynamics therapy combined with celecoxib for 24h, the protein expression of cyclooxygenase-2 was suppressed after, more compared with other three groups, the difference was significantly (P 〈 0.05 ). Conclusion Celecoxib could suppress proliferation of cervical carcinoma cells,and thestronger density, the obvious restrain. Photodynamics therapy combined with celecoxib can effectively inhibit the expression of target gene cyclooxygenase-2 and might 'be potentially useful in gene therapy of cyclooxygenase-2 related cervical carcinoma. Cyclooxygenase-2 gene can suppress the proliferation and promote the apoptosis in Hela cells.
