摘 要:目的检测乳腺癌患者组织和血清中血管内皮细胞生长因子(vascular endothelial growth factor,VEGF)、碱性成纤维细胞生长因子(basic fibroblast growth factor,bFGF)和组织中微血管密度(microvascular density,MVD)的表达,并观察其与乳腺癌生物学行为之间的关系,以及三者的相关性,探讨三者在乳腺癌的发生、发展、侵袭和转移中的作用。方法应用酶联免疫技术(ABC-ELISA方法)检测93例乳腺癌患者术前血清和20例健康体检者血清中的VEGF、bFGF表达水平,同时应用免疫组织化学染色方法检测该93例乳腺癌患者组织和20例乳腺腺瘤组织中VEGF、bFGF和CD34的表达。结果①术前未作放化疗的乳腺癌组患者术前血清VEGF、bFGF表达水平明显高于健康体检者、乳腺腺瘤患者,与原发肿瘤的大小、TNM分期、淋巴结转移有关(P<0.05);乳腺癌患者组织VEGF、bFGF阳性表达率、MVD值分别高于乳腺腺瘤患者,与原发肿瘤的大小、TNM分期、淋巴结转移有关(P<0.05)。②乳腺癌患者术前血清VEGF、bFGF的表达水平分别与组织VEGF、bFGF的阳性表达呈正相关(P<0.05);乳腺癌组患者组织VEGF和bFGF的阳性表达率呈正相关(P<0.01),血清VEGF和bFGF的表达水平呈正相关(P<0.05);乳腺癌患者术前血清与组织VEGF、bFGF表达水平分别与MVD值呈正相关。结论乳腺癌患者VEGF、bFGF及MVD在乳腺癌的发生、发展、转移中起着重要的作用,是乳腺癌术前诊断、判断转移危险的重要肿瘤标志物。Objective To investigate the expression and correlation of vascular endothelial growth factor(VEGF) , basic fibroblast growth factor(bFGF) and microvascular density(MVD) in human breast cancer and discuss their significance in carcinogenesis, progression, invasion and metastasis of breast cancer, as well as their relation with biological behaviour of breast cancer. Methods The expression of VEGF and bFGF were evaluated in the serum samples of 93 preoperation breast cancer patients as well as 20 serum samples of healthy persons using ABC-ELISA. The expression of VEGF, bFGF and CD34 were also studied in the tissues of breast cancer and normal tissue near cancer using immunohistochemistry. Results ① Theexpression levels of VEGF and bFGF in serum of preoperation breast cancer.patients without radiotherapy, chemotherapy were both significantly higher than those in healthy and breast adenoma persons, and were correlated with tumor size, TNM stage and the metastasis of lymph node (P 〈 O. 05 ). The expression rate of VEGF and bFGF and MVD in breast cancer tissues were higher than those in breast adenoma tissues respectively, and they were correlated with tumor size, TNM stage and the metastasis of lymph node (P 〈 0.05). ② The expression of VEGF and bFGF in serum of preoperation breast cancer patients was positively correlated with the expressions of VEGF and bFGF in breast cancer tissues respectively (P 〈 O. 05 ). The expression of VEGF in breast cancer tissues was positively correlated with the expression of bFGF respectively (P 〈0.01 ). The expression of VEGF in serum was positively correlated with the expression of bFGF respectively (P 〈 0.05 ). The expressions of VEGF and bFGF in serum of preoperation breast cancer patients and in breast cancer tissues was positively correlated with the MVD respectively. Conclusion VEGF, bFGF and MVD might play important roles in the development, progression, metastasis and prognosis of human breast cancer. They may be new markers for dia
