摘 要:目的观察非酒精性脂肪性肝病(NAFLD)大鼠血浆硫化氢(H2S)浓度变化。方法 SPF级雄性SD大鼠按随机数字法分为重度NAFLD模型组(重度组)、轻度NAFLD模型组(轻度组)、空白对照组(对照组)。重度组给予高脂饲料约20 g/d;轻度组高脂饲料量为重度组的50%,总重量与重度组相同;对照组给予重度组相同重量的普通饲料。12 w后每周处死1只重度组大鼠,观察脂肪肝程度;至16 w时,重度组达到重度脂肪肝,终止实验。腹主动脉采集血液,离心取血浆,检测甘油三酯(TG)、胆固醇(TC)、H2S及肝组织匀浆TG浓度。结果 (1)TC、TG:实验结束时,重度组TC、TG分别为(8.45±1.19)、(2.43±0.38)mmol/L,轻度组分别为(5.89±1.24)、(1.82±0.41)mmol/L,对照组分别为(1.98±0.14)、(0.84±0.22)mmol/L,3组间均有非常显著差异(P<0.01)。(2)肝组织TG:重度组(0.82±0.13)mmol/L,轻度组(0.62±0.14)mmol/L,对照组(0.42±0.10)mmol/L,3组间也均有非常显著差异(P<0.01)。(3)血浆H2S浓度:与对照组比较,重度组和轻度组H2S明显下降(P<0.01),重度组比轻度组下降更显著(P<0.05)。结论 NAFLD大鼠血浆H2S浓度下降,与脂肪肝程度相关;H2S下降是由于消耗过多。Objective To observe the concentration changes of hydrogen sulfide (H2S) in blood plasma of rats with nonalcoholic fatty liver disease (NAFLD). Methods Male Sprague Dawley ( SD ) rats at SPF level were randomly divided into model group of severe NAFLD ( severe group), model group of mild NAFLD ( mild group ), and blank control group ( control group). Severe group received high fat diet 20 g/d;mild group received high fat diet 10 g/d,and the total weight of diet was the same as that received by severe group;control group received normal feed 20 g/d. After 12 w, one rat in severe group was killed every week and its degree of fatty liver was observed. The experiment was ended in w 16 when the degree of fatty liver in severe group reached the severe level. Blood samples were collected from abdominal aorta and centrifuged to obtain plasma. The levels of plasma triglyceride(TG) ,cholesterol( TC), and H2 S and the TG concentration in liver tissue homogenate were detected. Results ( 1 ) After the end of the experiment, TC and TG levels in the severe group were respectively( 8.45 ± 1.19)and(2.43 ± 0.38)mmol/L;in the mild group were (5.89 ± 1.24)and (1.82± O. 41 ) mmol/L ; in the control group were ( 1.98 ± 0.14) and ( 0.84 ± 0.22 ) mmol/L. There were very significant differences among the three groups ( P 〈 0.01 ). (2) The TG level in liver tissues in the severe group, the mild group, and the control group were respectively(0. 82 ± O. 13 ) mmoi/L, (0.62 ± 0. 14 ) mmol/L, and (0.42 ± 0. 10 ) mmol/L, and there also were very significant differences among those groups ( P 〈 0. O1 ). ( 3 ) Compared with the concentration of H2S in plasma in the control group, the concentration in the severe group and the mild group decreased significantly ( P 〈 0.01 ), and it decreased more significantly in the severe group than that in the mild group ( P 〈 0.05 ).
