摘 要:目的:评价不同浓度过氧化脲对体外培养的人牙龈上皮细胞生长的影响,获得不同浓度过氧化脲引起人牙龈上皮细胞凋亡的时间范围。方法:体外培养人牙龈上皮细胞并进行鉴定,采用含1O%及20%过氧化脲的EpiLife上皮细胞专用无血清培养基进行培养,选取不同时间段计算细胞死亡率。结果:体外培养的人牙龈上皮细胞为多边形并呈铺路石状外观,角蛋白染色阳性,含10%过氧化脲培养基进行培养90s后细胞大量死亡,含20%过氧化脲培养基进行培养60s后细胞大量死亡。结论:过氧化脲对 体外培养的人牙龈上皮细胞具有较强的细胞毒性,临床常用过氧化脲污染牙龈时间不能超过1min。Objective To evaluate the effect of different concentrations of carbamide peroxide on the growth of cultured human gingival epithelial cells, and find out the time scope that apoptosis of the majority of the cells is induced during treatment with different concentrations of carbamide peroxide. Methods Human gingival epithelial cells were isolated and characterized by morphology and immunohistochemistry. The cells were then cultured in EpiLife medium in presence of 10% or 20% or in absence of carbamide peroxide. Percentage of dead cells was calculated at different time intervals upon carbamide peroxide supplementation. Results Cultured human gingival epithelial cells were polygonal, grew cobblestone-like in monolayer and stained positive for keratin. Death of the majority of the cells was observed within 90 seconds in the presence of 10% carbamide peroxide and within 60 seconds in the presence of 20% carbamide peroxide. Conclusion Carbamide peroxide is toxic to in vitro cultured human gingival epithelial cells. We suggest strongly that the time scope of carbamide peroxide contamination to gingiva should be well-controlled to be no more than 1 minute during clinical practice.
