摘  要:目的探讨趋化因子CXCL12及其受体CXCR4在人星形细胞瘤中的表达及意义。方法应用逆转录聚合酶链反应法(RT-PCR)、免疫组化SP法检测CXCL12和CXCR4在非瘤脑组织和人星形细胞瘤组织中的表达,分析CXCL12和CXCR4在不同病理级别人星形细胞瘤组织中的表达和它们之间的相关性。结果经统计学分析,CXCL12和CXCR4在非瘤脑组织和人星形细胞瘤组织中表达比较差异有统计学意义(P<0.05);在WHOⅠ~Ⅱ级人星形细胞瘤组织和WHOⅢ~Ⅳ级人星形细胞瘤组织中表达比较,差异有统计学意义(P<0.05)。CXCL12和CXCR4表达呈现明显的正相关(P<0.05)。结论 CXCL12和CX-CR4在人星细胞瘤中的表达高于在非瘤脑组织中的表达,其表达水平随病理级别的升高而升高,并且CXC L12和CXCR4之间存在明显的正相关。Objective To investigate the expression of chemokine CXCL12 and its receptor CXCR4 in human astrocytoma and its significances. Methods Reverse transcription polymerase chain reaction(RT-PCR) and immunohistochemistry SP were used to detected the expression of CXCL12 and CXCR4 in non-tumor brain tissues and human astrocytoma tissues,their expression in different pathological grades of human astrocytoma tissues and the correlation between CXCL12 and CXCR4 were analysed. Results By satistical analysis,the difference between the expression of CXCL12 and CXCR4 in non-tumor brain tissues and human astrocytoma tissues was statistically significant(P〈0.05), and the difference between the expression of CXCL12 and CXCR4 in WHO ] ~ H grades human astrocytoma tissues and WHO Ⅲ -Ⅳ grades human astrocytoma tissues was statistically significant(P〈0.05). The expression 6f CXCL12 and CXCR4 showed a significant positive correlation (P〈0.05). Conclusion The expression of CXCL12 and CXCR4 in human astroeytoma tissue is higher than the expression in non-tumor brain tissue,it increases with increasing patho logical grades and there is a significant positive correlation between CXCL12 and CXCR4.
