重度眼球穿孔伤视网膜光感受细胞损伤的实验研究

摘  要:目的观察兔重度眼球穿孔伤视网膜光感受细胞胀亡(oncosis)情况和核糖核酸(RNA)的表达,以及两者在重度眼球损伤中的作用。方法健康新西兰白兔制作重度眼球穿孔伤模型,用甲基绿-派罗宁-马休黄(MG.P-MY)染色法和超高倍镜分别观察细胞胀亡情况及进行细胞RNA检测。结果对照组和1h组未见胀亡细胞,3h组出现胀亡细胞,6h组胀亡细胞数达高峰,12h组有所下降。3h组在内核层及神经节细胞层有少量RNA阳性表达,6h内核层阳性表达增多,12h阳性表达明显,24h可见大量RNA阳性细胞。结论眼球枪弹穿孔伤后,胀亡是视网膜细胞死亡的另一种主要方式。在视网膜细胞死亡过程中,胀亡和凋亡是其主要死亡方式。Objective To investigate the oncosis cells and the RNA positive retinal photoreceptor cells after severe penetrating rabbit eye injury. Methods White New Zealand rabbits were selected. The RNA positive cell were observed through MG-P-MY stain methods, and the oncosis cell were observed under extremely high times microscopy. Results There was no oncosis cell in the control group or 1 hour experi- mental group. The oncosis cells could be seen in 3 hour group, more in 6 hour group, and the oncosis cell number decreased in 12 hour group. In 3 hour group, a lower positive expression of RNA were observed in the ganglion and inner layer cells, it increased obviously in 6 hour group, and there were more RNA expres- sion in 12 hour group. Numerous of RNA expression cells could be seen in 24 hour group. Conclusion The oncosis is the main way of the retinal cell dying after the rabbit penetrating eye injury. In the process of reti- nal cell death, oncosis and apoptosis is the main mode.
