摘  要:利用组织培养的方法培育香雪兰试管苗是获得脱毒种球的最佳途径之一。选取不带病毒的香雪兰茎尖作为外植体,通过诱导培养出试管苗,并进行试管苗成球研究。试验采用不同种类不同浓度的激素、活性炭的添加与否和2种培养温度进行3因素随机区组试验,共设3个重复,培养60d后,测定球茎诱导率和记录球茎质量。结果表明,香雪兰试管苗成球的数量和质量与激素的种类和浓度密切相关,并受到培养基中其他成分和环境温度的影响,通过数据分析,最佳培养条件是在MS培养基中添加NAA浓度为0.2mg/L、0.25%活性炭和6%蔗糖,光照条件为1 500lx,每天光照时间为14h,在低温13℃处理30d后再在25℃条件下处理30d。Culturing tube seedlings of Freesia refracta in vitro is one of the best methods for obtaining virus-flee corms. The virus--flee shoot tips were selected as explants and new seedlings were cultivated, and the process of corms formation from seedlings was conducted. Three-factor Randomized Blocks test with 3 repeat was performed with different variety and concentration of hormones, with or without activated carbon and two temperature conditions. After the tube seedlings were inducted for 60 days in vitro, the number of bulbs and the weight of each corm were statistically analyzed. The results showed that the quantity and quality of the produced bulbs were closely related with the species and concentration of the used hormone, and effected by the component in culture medium and the environmental temperature. The data analysis indicated that the proper culturing conditions for tube seedlings of Freesia refracta in vitro were as follows: MS medium with 0. 2 mg·L^-1 NAA, 0. 25% activated carbon and 6% sucrose, illumination with 1500 LUX and 14 hours a day, 30 days with 13 ℃ and 30 days with 25 ℃.
