摘 要:纤维素酶可提高饲料利用率,对于畜牧养殖具有重要意义。本文利用实验室分离获得的纤维素降解活性较高的菌种Bacillus.sp.A-1-3,采用盐析法对纤维素酶进行提取纯化,对影响酶液提取的主要因素进行了正交试验,优化了纤维素酶的提取条件。实验结果表明:纤维素酶的最佳提取条件是pH值4.4,温度50℃,硫酸铵饱和度为70%,提取时间为12h,此时的酶活为36.772U/ml。Cellulase,an addition that credited as feed enhancer,play very important role in animal feeding industry.In this research,a laboratory isolated strain Bacillus.sp.A-1-3 with potent cellulase catalytic activity was applied as experimental substance.The concerned cellulase were purified by salting out method.Based on orthogonal design,factors that affect the purification of cellulase solute were optimized by testing any necessary combinations.The result indicates that best cellulase purification can be expected when pH was sett at 4.4,temperature was stabled at 50°C,(NH4)2SO4 was saturated by 70%,time duration of extraction was controlled at about 12 hour.In this setting,cellulase enzyme activity was about 36.772U/ml.
