β_2-ADR27位点基因多态性与胰腺癌相关性

摘　　要：目的探讨β2肾上腺素能受体(β2-ADR)27位点基因多态性与胰腺癌的相关性。方法应用聚合酶链反应-限制性片段长度多态性(PCR-RFLP)方法结合DNA测序法检测32例胰腺癌组织和10例正常胰腺组织中β2-ADR 27位点基因多态性,分析其多态性与临床病理特征之间的关系。结果 27位点CC、CG基因型分布频率在胰腺癌组分别为68.75%、31.25%,在正常胰腺组为80.00%、20.00%,两组间差异无统计学意义(P>0.05)。C和G等位基因频率在胰腺癌组为84.38%和15.62%,在正常胰腺组为90.00%和10.00%,两组间差异无统计学意义(P>0.05)。27位点基因多态性与病人的性别、年龄、肿瘤分化程度、肿瘤大小、淋巴结是否转移和TNM分期均无相关性(P>0.05)。结论β2-ADR 27位点基因多态性可能与胰腺癌发病无相关性,尚需大样本的进一步研究。Objective To investigate the correlation between the single nucleotide polymorphism （SNPS） in position 27 of β2-adrenergic receptor （β2-ADR） and pancreatic carcinoma （PC）. Methods By using PCR-restriction fragment length poly- morphism （PCR-RFLP） and DNA sequencing, the polymorphism in position 27 of β2-ADR in 32 cases of PC and 10 healthy con- trols were detected. The correlation between the polymorphism and clinicopathologic characters of PC was analyzed. Results The distribution frequency of two genotypes, CC and CG, in position 27 in PC was 68.75%and 31.25%, respectively, in normal pancreatic tissue, that was 80.00% and 20.00%, the differences between the two groups were not significant （P〉0.05）. The fre- quency of C and G allele was 84.38% and 15.62% in PC, and 90.00% and 10.00% in normal pancreas, with no statistical differ- ence between the two groups （P〉0.05）. The SNPS in position 27 bad no significant correlation with the gender, age, histological differentiation, size of tumor, lymph node metastasis and TNM staging （P〉0.05）. Conclusion There might be no correlation between the polymorphism in position 27 of β2- ADR and the development of pancreatic carcinoma, more further large-sample stud- ies are needed.
