摘 要:目的 :应用分离的骨髓干细胞 (MSC)进行牙周骨缺损的自体移植动物实验 ,观察MSC用于牙周再生治疗的可行性。方法 :体外分离狗髂骨的MSC ,体外培养 ,移植前进行MSC的体外钙化实验 ,以胶原膜为移植载体 ,将MSC移植到狗下颌后牙的根分叉骨缺损中 ,覆盖国产聚四氟乙烯膜 ,30d后进行组织学观察。结果 :分离的MSC在体外均出现了不同程度的钙化 ;动物实验结果发现 ,MSC移植组达到了骨缺损的完全修复 ,无根面吸收 ;对照组骨再生高度明显低于实验组 (P <0 .0 1) ,并有上皮结缔组织长入。两组均有明显的新生牙骨质的形成。结论 :MSC移植可加快牙槽骨的再生 ,治疗牙周骨缺损的研究和应用是可行的。AIM: To evaluate the effect of transplantation of bone marrow stem cells on regeneration of periodontal bone defect. METHODS: MSC was isolated from iliac marrow of two dogs. Cells were cultured and mineralized in vitro treated with dexamethasone, -Glycerophosphate and ascorbic acid.In the mean time, cells were transplanted into the furcation bone defects using collegen carrier and e-pTFE membrane were covered. The histological observation was carried out after 30 days. RESULTS: MSC showed the mineralized character in vitro. The bone defects transplanted with MSC were fully restored, and the defects in control group only covered with e-pTFE membrane were partially restored. Epithelium and connective tissue between furcation and top of new bone were observed in control group. New cementum was formed in both groups. CONCLUSION: The periodontal bone defect of furcation can be fully repaired with MSC transplantation without epithelium and connective growed into. The results indicated that MSC was hopeful cell source of transplantation for periodontal bone defect.
