摘 要:目的:利用流式细胞术外周血微核试验方法,研究环磷酰胺(cyclophosphamide,CP)和甲磺酸乙酯(ethyl methanesulfonate,EMS)致大鼠外周血微核的发生率。方法:对SD雄性大鼠分别给予不同剂量的CP和EMS处理,在给药前和给药后24,48,72和96 h实施尾静脉采血,对血样进行固定、转移、洗脱、标记和染色后,利用流式细胞仪对样本进行测定。计数20 000个网织红细胞中含微核细胞的数量,计算微核细胞百分率和网织红细胞占全血的比率,并对数据进行统计分析。结果:CP和EMS均可诱发外周血网织红细胞比率下降和微核细胞率增加,伴随时间延长两项指标均可恢复至给药前水平,给药后48 h是检测微核的最佳时间。结论:CP和EMS可以对大鼠外周血网织红细胞微核率产生显著影响。Objective: To analyze the mieronuclei in peripheral blood retieulocytes of rats induced by cy- clophosphamide (CP) and ethyl methanesulfonate (EMS) with flow eytometry. Methods: Male SD rats were trea- ted with various doses of CP and EMS. Sample blood was collected from tail vein at predose and at 24, 48, 72 and 96 h after dosing. The blood was dealt with following processes: fixing, transferring, eluting, labeling and dyeing. The sample was detected with flow cytometry to count micronucleated cells in 20 000 reticulocytes. Micronucleated reticulocytes percentage and retieulocytes percentage in blood were calculated. Results: CP and EMS decreased re- ticulocyte rate and increased micronucleated rate in peripheral blood. At 48 h after dosing, detection of micronuclei was the most sensitive. Conclusion: CP and EMS can significantly affect the mieronucleated rate of retieulocytes.
