原发性结肠癌组织RUNX3基因表达及临床意义

摘　　要：目的检测原发性结肠癌组织RUNX3基因mRNA和蛋白质表达并探讨其与临床病理因素之间的关系。方法采用RT-PCR和免疫组织化学法检测101例原发性结肠癌组织和正常结肠组织中RUNX3基因表达，分析RUNX3基因表达与临床病理因素间的关系。结果82例（81．2％）RUNX3基因tuRNA呈无表达或低表达状态，对应的蛋白质表达呈低表达或无表达状态，而正常结肠组织中mRNA和蛋白质表达正常；随着Dukes分期越晚，RUNX3基因mRNA表达越少。结肠癌组织中RUNX3基因表达在性别与年龄中的差异无统计学意义（P〉0．05）；随着病理分型恶性程度增加，RUNX3基因表达减少（P〈0．05）；Dukes分期越晚，结肠癌组织RUNX3基因表达越低（P〈0．05）。结论RUNX3基因表达在原发性结肠癌组织中显著降低，对结肠癌临床分期和病理诊断有重要临床意义。Objective To study the expressions and clinical significances of RUNX3 gene in primary colon cancer. Methods RT-PCR and immunohistochemistry were used to detect the expressions of RUNX3 gene in 101 cases with primary colon cancer. Results 82 cases （81.2%） showed low or no mRNA expression of RUNX3 gene with corresponding protein expression, while it＇ s normal in normal colon tissues. The mRNA expressions lower down as Dukes staging＇ s increase. The gene expression of RUNX3 decreased with pathylogical malignance and Dukes staging＇ s increase. Conclusion The expressions of RUNX3 gene in primary colon cancer are low, which have significant clinical significances for colon cancer＇ s staging and diagnosis.
