摘  要:目的研究不同浓度的喷他脒对卵巢癌SKOV3细胞的迁徙侵袭作用的影响。方法以空白卵巢癌SKOV3细胞为对照组,以不同浓度喷他脒作用于卵巢癌SKOV3细胞作为实验组,即0.10、1.00、10.00和100.00μmol/L组,以Transwell小室法培养各组SKOV3细胞,采用MTT法分别检测上、下室细胞在波长450 nm处的OD值,以细胞的侵袭率和迁徙率来表示肿瘤细胞的侵袭、迁徙能力。结果喷他脒的高、中浓度组抑制卵巢癌SKOV3细胞的迁徙能力和侵袭能力有显著差异。结论不同浓度的喷他脒可以抑制卵巢癌SKOV3细胞的迁徙能力和侵袭能力,并呈明显的浓度依赖关系,低浓度组喷他脒抑制迁徙能力较弱,高浓度组抑制作用最强。Objective To investigate the inhibiting effect of different dose pentamidine on the metastatic invasion of ovarian cancer cells SKOV3. Methods The blank ovarian cancer cells SKOV3 were used as the control group and different concentrations of pentamidine in ovarian carcinoma cells SKOV3 were used as the experimental group. The Transwell method was applied to culture the SKOV3 cells of the 3 groups. The OD values at the wavelength of 450 nm were measured with MTT method. The invasive and migrating rates of the ceils were applied to indicate the invasive and migrating capacity of tumor cells. Results Experiments showed that significant differences could be seen in the migrating and invasive capacity of SKOV3 cells for the high, medium and low pentamidine dosage groups. Conclusion Different concetrationss of pentamidine could inhibit the migrating and invasive eapacity of SKOV3 cells to various extents, and a dosage effect relationship could clearly be seen: lower dosage of pentamidine had a weaker inhibitive effect, while the higher dosage of pentamidine had the strongest inhibitive effect.
