摘　　要：目的研究Trophinin(Tro)基因在胰腺癌组织中的表达及临床意义。方法选择手术切除的胰腺癌组织及癌旁组织标本32例,正常胰腺组织10例。免疫组化SP法检测Tro蛋白的表达水平;追踪随访患者术后24个月生存状况,并与实验结合进行统计学处理及结果分析。结果免疫组化Tro蛋白主要表达在癌细胞细胞核、核膜,少量表达在细胞质。Tro在胰腺癌组织、癌旁组织及正常胰腺组织中的表达阳性率分别为81.3%(26/32)、40.6%(13/32)、10.0%(1/10)。Tro蛋白的表达与患者的性别、年龄、肿瘤的部位、CEA、CA199以及CA125无关;与肿瘤临床分期、肿瘤的分化程度以及神经侵袭关系密切。追踪随访患者的术后生存期:Tro高度表达的患者24个月生存期明显低于低度表达及阴性表达的患者。结论 Tro在胰腺癌的发生、发展中可能起重要作用;Tro高表达者预后差,Tro可以作为胰腺癌临床预后判断的分子指标。Objective To investigate the expression and clinical significance of trophinin gene in pancreatic cancer.Methods 32 surgery resected pancreatic cancer and adjacent tissue specimens,and 10 normal pancreatic tissue were selected.SP immunohistochemical staining was used to measure the expression levels of trophinin protein.Patients after operation were followed up for 24 months,and the data was analyzed in conjunction with experimental results and statistical analysis.Results Trophinin protein was mainly expressed in nucleus and nuclear membrane of pancreatic cancer cells,small amount was expressed in the cytoplasm.Trophinin protein expression in pancreatic tissues was 81.3%（26/32）,in adjacent noncancerous tissues 40.6%（13/32）,in normal pancreatic tissue 10.0%（1/10）.Trophinin protein expression was not related with patients′gender,age,tumor location,CEA,CA199 and CA125;however it was closely correlated with clinical stage,tumor differentiation and neural invasion.Follow up of patients survival：the survival of patients with high expression of Trophinin within 24 months was significantly less than those with moderate,low and negative expression of Trophinin.Conclusion Trophinin plays an important role in the genesis and development of pancreatic cancer.Strong Trophinin expression indicates poor prognosis,which can be used asa molecular markderof pancreatic cancer prognosis.
