摘  要:目的观察芦根多糖对免疫性肝纤维化大鼠的保肝及抗肝纤维化作用,探讨其抗肝纤维化的机制。方法Wistar大鼠随机分为正常组、模型组、阳性药组及芦根多糖高、中、低剂量组。除正常组外,腹腔注射猪血清诱导大鼠免疫性肝纤维化模型,造模7周后各给药组开始灌胃给予相应药物,正常组及模型组给予等量生理盐水。检测血清天冬氨酸氨基转移酶(AST)、丙氨酸氨基转移酶(ALT)、超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-px)活性,丙二醛(MDA)含量及肝组织SOD、GSH-px活力和MDA含量。结果芦根多糖可降低模型大鼠血清AST、ALT活性,降低血清和肝组织MDA含量,升高血清和肝组织SOD、GSH-px活力。结论芦根多糖可改善肝纤维化模型大鼠肝功能,减轻纤维化,其机制可能与其保肝降酶、抗脂质过氧化有关。Objective To observe the effect of polysaccharide of Rhizoma Phragmitis(PRP) on immune hepatic fibrosis model rats induced by porcine serum and to explore the mechanism of protecting liver and anti-hepatic fibrosis.Methods The Wistar rats were randomly divided into six groups:normal group,model group,control group,and high dose(420 mg/kg),medium dose(210 mg/kg) and low dose(105 mg/kg) treatment groups of PRP.Immune liver fibrosis model were established by injecting porcine serum 0.5 mL per time per rat for 15 weeks,twice per week except the normal group.Rats were intragastrically administrated for 8 weeks successively from the 7th week to the 15th week.ALT,AST,SOD,MDA,GSH-px in serum and SOD,MDA,GSH-px in tissue of liver were detected.Results PRP could decrease activity of ALT and AST,increase the activity of SOD and GSH-px in serum and in tissue of liver,decrease the level of MDA in serum and in liver tissue.Conclusion PRP can protecting liver function of hepatic fibrosis rats induced by porcine serum.The mechanism of anti-fibrosis may be associated with decreasing the activity of enzyme and anti-oxidation.
