摘 要:目的探讨氟-1 8标记脱氧葡萄糖(^(18)F-FDG)微正电子发射型计算机断层扫描/CT扫描(Micro-PET/CT)对^(125)Ⅰ放射性粒子组织间植入治疗人胰腺癌裸鼠移植瘤模型的评估价值。方法将人胰腺癌SW1990细胞株接种于18只BALB/C裸鼠.并将荷瘤裸鼠随机分为空白对照组、空载粒子组和^(125)Ⅰ粒子植入组,每组6只。分别于治疗前及治疗后1周行^(18)F-FDG Micro-PET/CT检查,计算最大标准摄取值(SUVmax)、平均标准摄取值(SUVmean)。瘤体标本进行胸苷激酶1(TK1)检测细胞增殖及末端脱氧核苷酸转移酶-生物素dUTP切口末端标记法(TUNEL)检测细胞凋亡。结果治疗前三组间SUVmax和SUVmean值差异无统计学意义(P>0.05)。治疗后1周,^(125)Ⅰ粒子植入组SUVmax和SUVmean分别为0.28±0.20和0.02±0.01,明显低于空白对照组分别为(3.54±1.49和0.59±0.18)和空载粒子组(分别为3.78±1.67和0.57±0.21).三组间SUVmax和SUVmean值差异有统计学意义(P<0.05)。^(125)Ⅰ粒子植入组TK1阳性染色指数为37.86±1.71,明显低于空白对照组(64.01±1.48)和空载粒子组(62.88±2.36),差异有统计学意义(P<0.01);而^(125)Ⅰ粒子植入组的凋亡指数为23.78±2.53,明显高于空白对照组(7.99±1.32)和空载粒子组(8.36±1.60),差异亦有统计学意义(P<0.01)。SUVmax与TK1阳性染色指数呈正相关,而与凋亡指数呈负相关。结论 ^(18)F-FDG Micro-PET/CT能有效监测胰腺癌近距离放射治疗的疗效,是一个有效的评估手段。Objective To discuss the value of 125F-FDG Micro-positron emission tomography (PET)/ computed tomography (CT) in assessing the early therapeutic effect of 125I interstitial brachytherapy on transplantation tumor of human pancreatic carcinoma in Balb/c-nu mice. Methods Xenograft models were established by subcutaneously injecting SW1990 human pancreatic cancer cell suspension into the immuno- deficient Balb/c-nu mice. A total of 18 experimental mice were randomly and equally divided into three groups: control group (n = 6), ghost seed implant group (n = 6) and 125I seed implant group (n = 6). Before treatment and one week after treatment, 18F-FDG Micro-PET/CT scanning was performed, and then maximum standardized uptake values (SUVmax), mean standardized uptake values (SUVmean), tumor size and necrosis rate were measured. Cell proliferation and apoptosis were monitored in vivo by thymidine kinase 1 (TK1) immunostaining and Dutp-biotin nick end labeling (TUNEL) assay respectively. Results Before the treatment no statistically significant d/fference in SUVmax and SUVmean values existed between each other ofthe three groups. One week after the treaLrnent the differences in SUVmax and SUVmean values between each other of the three groups were statistically significant (P 〈 0.05). In 125I seed implant group the SUVmax and SUVmean values became significantlylower than those determined before the treatment, while in ghost seed implant group and control group the SUVmax and SUVmean values showed no obvious changes when compared to the values obtained before the treatment. analysis of the tumor tissues revealed that TK1 cell positive staining index of 125I seed implant group was (37.86 ± 1.71), which was significantly lower than that of control group (64.01 ± 1.48) and that of ghost seed implant group (62.88 ± 2.36), and the differences were statistically significant (P 〈 0.01).
