摘  要:目的:考察淫羊藿甙(ICA)对甲状腺癌细胞系SW579的增殖、细胞周期、黏附及侵袭效应的影响,探讨其逆转甲状腺癌细胞系SW579恶性表型的机制。方法:采用MTT法检测ICA作用过的甲状腺癌细胞系SW579的增殖,运用流式细胞技术(FACS)检测细胞周期分布;运用Tanswell小室侵袭试验,软琼脂集落形成实验,细胞-基质黏附率检测等方法检测逆转SW579恶性表型情况;RT-PCR检测分化抑制因子/DNA结合抑制因子(Id-1)、p21 mRNA表达水平。结果:与对照组比较,ICA在浓度(6.25~100)mg/L范围内,对SW579细胞增殖的抑制作用呈明显的浓度依赖效应和时间依赖效应;细胞周期各时相分布中S期明显减小(P<0.01),而G0/G1期升高(P<0.01);SW579细胞非整倍体DNA含量减少,克隆形成率、细胞基质黏附率以及体外细胞侵袭力明显降低;Id-1 mRNA表达下调,p21mRNA表达升高,且有明显的浓度依赖性。结论:ICA可以逆转人甲状腺癌SW579细胞系的恶性表型,通过下调Id-1 mRNA水平来激活p21基因的表达,从而使SW579细胞从S期向G1/G0期逆转,完成诱导分化。Objective:To observe the effect of icariin(ICA) on cell proliferation,cell cycle,adheration and invasion in SW579 cells and to approach the mechanism of its reversing the malignant phenotype of SW579 cells.Methods:MTT assay was used to explore theeffects of ICA on the proliferation of SW579cells.Reverse transcriptase-polymerase chain reaction was used to examine mRNA levels of inhibitor of differentiation/DNA binding-1(Id-1)and p21 genes.Flow cytometry was used to detect cell cycle of SW579 cells treated by IP.Soft agar colony formation was used to test the anchoring independent growth of SW579 cells.Adhesion between SW579 cells and matrigel was be examined and the Transwell chamber was used to detect the invasion.Results:The inhibition of SW579 cells proliferation treated by ICA was time and concentration dependent in the the concentration range 6.25 mg/L^100 mg/L.The cell cycle status of ICA-treated SW579 cells were significantly different compared with those of the controls.The number of G0/G1-phase cells increased(P=0.0025),and S-phase cells decreased(P=0.001),and the aneuploid content decreased simultaneously.The colony forming efficiency,adhesion rate between SW579 cells and matrigel and invasiveness were reduced remarkably.ICA down-regulated the expression of Id-1and up-regulated the expression level of the downstream effector p21 mRNA.Conclusion:Icariin reversed the malignant phenotype of sw579 cells and induced the differentiation of SW579 cells through down-regulated Id-1,which activate the p21.
