摘  要:目的探讨替莫唑胺对大鼠垂体腺瘤GH3细胞增殖、凋亡及其泌乳素分泌的影响。方法采用不同浓度的替莫唑胺作用于体外培养的GH3细胞(以加入DMSO稀释液的细胞为对照组),分别作用24、487、2、96 h后,采用MTT比色法检测细胞增殖,原位末端标记(TUNEL)法检测细胞凋亡,ELISA法检测细胞合成分泌泌乳素情况。结果替莫唑胺在体外可明显抑制GH3细胞增殖,具有浓度、时间依赖性(P<0.05);加入替莫唑胺后的细胞凋亡率均明显高于对照组(P<0.05);替莫唑胺作用24 h后,各时间点泌乳素浓度均低于对照组(P均<0.05)。结论替莫唑胺可显著抑制GH3细胞增殖并促进其凋亡,且抑制其泌乳素的合成分泌。替莫唑胺可尝试用于难治性垂体腺瘤的治疗。objective To evaluate the effect of temozolomide (TMZ) on GH3 pituirary ademomas cell proliferation, apoptisis and hormonal production. Methods The different dose of TMZ was added to GH3 cells cultured in vitro, the control group was added with DMSO diluent, after administration of 24, 48, 72, 96 h, cell proliferation was assessed by MTT assay, apoptosis was detected, by TUNEL staining, the change of prolactin secretion was measured by ELISA. Results TMZ could inhibit GH3 cell proliferation which depended on the time and concentration of treatment significantly( P 〈 0.05 ). GH3 cell apoptosis with added TMZ was significantly higher compared to control group ( P 〈 0.05 ). After 24 h administration of TMZ, the prolaetin secretion of GH3 cell was lower compared to control group. Conclusions TMZ can inhibit GH3 pituirary ademomas cell proliferation and induce apoptosis and supress hormonal secretion, these benefits prove that TMZ could be used in the management of aggressive pituitary ademoma treatment.
