摘  要:目的了解胃癌细胞系SGC7901在缺氧状态下凋亡和增殖情况,探讨低氧诱导因子(hypoxia induced factor,HIF-1α)与胃癌细胞SGC7901凋亡和增殖的关系。方法以二氯化钴(CoCl2)作为缺氧模拟剂,利用RT-PCR、免疫荧光及Western blotting法检测SGC7901细胞HIF-1α在转录水平和翻译水平上的表达,利用流式细胞仪检测SGC7901细胞在不同缺氧程度下的细胞凋亡率,利用MTT法观察SGC7901细胞的增殖情况。结果 (1)50μM CoCl2处理SGC7901细胞24小时后,HIF-1a mRNA及蛋白表达明显增加;(2)50μM CoCl2处理SGC7901细胞24小时后,细胞凋亡减少;(3)50μM CoCl2处理24小时后,细胞生长和增殖均加快。结论 50μM CoCl2处理SGC7901细胞24小时后,细胞增殖能力增加,细胞凋亡减少,HIF-1α表达增加,提示其在此过程中发挥重要作用。Objective To investigate the apoptosis and proliferation of gastric carcinoma cell line SGC7901 induced by hypoxia,and to study the role of HIF-1α in the the apoptosis and proliferation course of gastric carcinoma cells.Methods SGC7901 cells were treated with 50 μM CoCl2 for 24 hours.The expressions of HIF-1α at the levels of transcription and translation were detected by RT-PCR,immunofluorescence and Western blotting;and the change of cell apoptotic ratio were detected by flow cytometry;and the change of cell proliferation were detected by MTT.Results(1)HIF-1α was upregulated both at the transcriptional and translational levels under hypoxic condition;(2)apoptotic ratios of SGC7901 cells were decrease;(3)Proliferation of SGC7901 were increase.Conclusion 50 μM Cobalt dichloride can decrease apoptosis and induce proliferation of gastric carcinoma cell line SGC7901,and HIF-1α plays an important role in this process.
