黄芪建中汤对脾气虚肺癌小鼠免疫功能及cyclin D_1基因表达的影响

摘　　要：目的观察黄芪建中汤对脾气虚肺癌小鼠免疫功能及cyclin D1基因表达的影响,探讨黄芪建中汤对脾气虚肺癌小鼠抑瘤作用的可能机制。方法用苦寒泻下法建立小鼠脾气虚模型,并接种Lewis肺癌瘤株。造模小鼠随机分成正常组、肿瘤对照组、模型对照组、黄芪建中汤低、中、高剂量组,用药14 d后,眼球取血,检测血清木糖及胃泌素水平;取肿瘤组织,用RT-PCR方法检测cyclin D1-mRNA表达。结果黄芪建中汤明显改善脾气虚小鼠的整体状况;抑制肿瘤的生长,提高小鼠脾指数、胸腺指数;提高脾气虚小鼠的血清木糖、胃泌素;下调肿瘤组织cyclin D1-mRNA的表达。结论黄芪建中汤可能是通过改善脾气虚肺癌小鼠的免疫水平、下调肿瘤组织cyclin D1-mRNA的表达,达到抑制肿瘤的生长。Objective： To observe the effects of Huangqijianzhong Decoction on immune functions and cyclin D1 gene expression in mice of lung cancer with spleen-qi deficiency to discuss its anti-tumor effect and molecular mechanism preliminarily.Methods： Spleen-qi deficiency model was induced in mice by radix and vaccination lewis lung cancer tumors strains.These mice were divided randomly into the normal control group,the tumor control group,the model control group,the low dose of Huangqijianzhong Decoction group,the middle dose of Huangqijianzhong Decoction group and the high dose of Huangqijianzhong Decoction group.After 14 days,some indices were observed and compared in above groups such as the serum xylose concentration,the gastrin level and the cyclin D1-mRNA expression.Results： The whole situations of mice with spleen qi deficiency syndrome and the growth conditions of lung cancer were improved by the Huangqijianzhong Decoction.Some indicators were increased by this decoction including the spleen index,the thymus index,the serum xylose and the serum gastrin.However,the cyclin D1-mRNA expression was inhibited by Huangqijianzhong Decoction.Conclusion： The mechanism of that Huangqijianzhong Decoction inhibits the growth of tumors may be improving the immune level and regulating down the cyclin D1-mRNA expressions in mice with spleen-qi deficiency syndrome.
