摘 要:目的 :提高肿瘤细胞的免疫原性。方法 :将异基因型或同基因型的MHCⅡ类基因转染至小鼠肥大细胞瘤P815中 ,用此细胞接种同系小鼠 ,1个月后给不成瘤的小鼠再接种野生型P815细胞 ,观察小鼠的存活情况。结果 :转同基因型MHCⅡ基因的P815细胞接种同系小鼠后 ,4/ 10不成瘤 ,而异基因型MHCⅡ基因转染组 ,10 / 10不成瘤。 1个月后给不成瘤的小鼠再接种野生型的P815细胞 ,同基因型转染组 2 / 4不成瘤 ,异基因型转染组 7/ 10不成瘤。结论 :研究说明MHCⅡ基因转染具有使肿瘤细胞的致瘤性下降、免疫保护能力提高的作用 ;并且异基因型比同基因型MHCⅡ类基因转染的效果更佳。Objective:To enhance the immunogenicity of tumor cells.Methods:Allogeneic or syngeneic MHC class Ⅱ gene was transfected into murine mastocytoma P815.Then the transfected cells were inoculated into the syngeneic mice and one month later ,mice free of tumors were challenged with wild type P815 cells.The survival rate of the mice was observed.Results:4/10 mice were free of tumors when inoculated with syngeneic MHC class Ⅱ gene modified P815 cells,while 10/10 mice were free of tumors when inoculated with allogeneic one.One month later,the mice free of tumors were challenged with wild type P815 cell and the survival rate of syngeneic group was 2/4,while the allogeneic one was 7/10.Conclusion:Expression of MHC class Ⅱ molecular remarkably reduced the tumorigenicity of the modified cells and vaccination with MHC class Ⅱ expressing P815 cells protected the mice from subsequent challenge with wild type P815 cells.Furthermore,allogeneic MHC class Ⅱ transfection was better than syngeneic one in their ability to generate immune protection effects.
