摘 要:细胞增殖是可调控的 ,细胞周期正向和负向调节因素参与其中。目前已发现细胞周期蛋白 /周期蛋白激酶抑制剂同细胞周期G1期的调控有关。这些抑制剂中 ,p2 7kip1的研究特别受重视 ,在G1期细胞周期蛋白 /周期蛋白激酶复合物的调节中起主要作用。p2 7高表达可引起G1期阻滞 ,抑制多种癌细胞的生长。目前仅在少数种类的肿瘤中检测了p2 7的免疫活性。在结肠癌和乳腺癌中 ,p2 7低表达与肿瘤进展和低生存率相关。研究证明p2Cell proliferation is governed by the synthesis and activation of a number of positive and negative regulators of cell cycle progression. A number of cyclin/CDK inhibitors have been found associated with the G1 cell cycle period. One of these inhibitors,p27 kip1 , has been identified by a number of investigators independently and appears to play a major role in the regulation of cyclin CDK complex activity in this phase of the cell cycle. Overexpression of p27 in mammalian cells induces a G1 block of the cell cycle and inhibits growth of a number of cancer cells. p27 immunoreactivity,however,has been investigated in only a limited number of human cancers. Low p27 expression in colon cancer and breast cancer is associated with tumor progression and decreased survival. p27 expression has been found to be an independent prognostic marker.
