摘 要:运用多聚酶链扩增反应(PCR)及瑞粒重复扩增方法(TRAP)法检测肿瘤细胞Hela、A431和KB在电离辐射后hEST2功能区片段表达和瑞粒酶活性的变化。结果表明:肿瘤细胞hEST2功能区片段表达和端粒酶活性呈剂量依赖性下降;hEST2功能区片段的PCR法测定结果与TRAP法相符合;提示电离辐射对瑞粒酶活性的抑制是肿瘤放射治疗的重要机制之一。端粒酶活性检则可能成为评估电离辐射对肿瘤杀伤作用的新指标。Telomerase has been detected in more than 85% of human tumor cell lines and tissure,but not exist in majority of nomal cell and tissure. PCR and TRAP were employed in this experiment to test the telomerase activity changing of tumor cell line Hela,A431 and KB after irradiated.The result indicated that:tumor telomerase activity and expression of hEST2 functional area fragment are dose-dependent decreased;the result of PCR test method are corrlated with that of TRAP,those data suggest that the supression effect of irradiation onto tumor telomerase activity is one of critical mechanism of radiotherapy.Determining the telomerase activity level perhapes is a new index to evaluate the killing effect of irradiation onto tumor.
