摘  要:目的:建立荧光活性染料CFSE以适宜终浓度标记体外培养的嗅鞘细胞的染色方法。方法:CFSE以2、5、10μM 3种不同终浓度标记原代培养的嗅鞘细胞,通过流式细胞术、台盼蓝染色和荧光显微镜,检测和观察CFSE染色的阳性细胞率、细胞活力、生长形态特点和增殖情况。结果:3种终浓度CFSE标记嗅鞘细胞的阳性标记率均>99%;2μM CFSE标记的细胞活力最高(P<0.05),细胞生长增殖较快,但随时间推移荧光强度明显减弱。结论:CFSE可用于标记嗅鞘细胞,本研究条件下,2μM的终浓度较为适宜。Objective: To establish a method of CFSE labeling olfactory ensheathing cells (OECs) at appropriate final concentration in vitro. Methods: OECs were labeled by 2, 5, or 10 μM CFSE in vitro. The positive labeling rate, cell viability, growth and morphological charac- teristics and proliferation of OECs were detected by flow cytometry, trypan blue staining and fluorescence microscopy respectively. Results: More than 99% of the OECs were CFSE-labeled at the three concentrations (2, 5, 10μM) investigated. The OECs labeled with 2μM CFSE had the highest viability and the maximum proliferation rate. However, the fluorescence intensity was significantly decreased with time. Conclusion: CFSE can be used to label OECs with optimized concentration of 2 μM.
